How should fiber diameter statistics and images be reviewed?

The mean describes average diameter; SD and CV describe dispersion among the objects in that report. These statistics do not directly establish instrument accuracy or repeatability. Check effective counts, images, markings, and statistical rules.

Confirm the measured objects

Check material, preparation, model, scan region, and the actual effective count. Support for 10,000-fiber tasks does not mean every quick scan must measure 10,000 fibers.

Mean, SD, and CV

The mean describes the center of the distribution. SD describes dispersion, and CV is usually SD divided by the mean. Variation among fibers and variation between repeated measurements are different questions.

Percentiles are not proportion labels

D10, D50, and D90 are percentile values of a cumulative distribution. Confirm whether the variable is diameter or length and what statistical weighting is used. D10 should not simply be called the proportion of fine fibers; diameter and length variables must remain distinct.

Review the images

Inspect markings on edge objects, crossings, curved fibers, contamination, and overlaps. Review exclusions with the software version and actual sample. Screenshots illustrate the process; quantitative claims require complete test conditions.

Review counts before distributions

Video 37 shows included and excluded objects, source images, and measurement lines. After an authorized redraw or exclusion, review the updated effective count and report using the same revision.

Paired comparisons and whole-slide statistics

In video 71, manual and automated means for the same 25 fibers are 8.745 and 8.928 μm, a difference of 0.183 μm. A paired comparison addresses method differences on the same objects; whole-slide statistics describe a larger effective population. Neither should be combined into an accuracy claim.

Rescanning and independent preparation

Video 71 shows means of 9.17–9.18 μm across five reports from one slide, with different effective counts. Video 52 compares independently prepared slides: 2,137 fibers at 11.84 μm and 1,903 at 11.90 μm. The latter also includes preparation and sampling differences. A difference between means is not the complete instrument accuracy.
